A Smoke Test is a very basic level test of a program or new/repaired hardware that serves as an indicator of whether the rudimentary or most crucial functions are working.
This could be as simple as seeing whether a new or freshly repaired computer will turn on at all without incident (i.e. smoke coming out of it literally), or seeing if a newly written program will at least run in some manner (even though it may not do much else at the moment).
Once a program or piece of hardware has passed the Smoke Test stage, then the work on fine tuning, tweaking, and/or intricate details can begin.
