Does any know if disabling this really increase boot time?

I'm not sure but I wouldn't recommend it.
The LUAFV driver handles User Account Control (UAC) File Virtualization.
A feature that redirects writes to protected locations to your local virtual store folder to allow some older applications to run with UAC on without needing Administrative privileges.

I have turned UAC off, would it still affect my laptop by turning it off?

I have no idea. I usually leave Vista system services alone if possible.
There can be unexpected side effects from disabling them.
