Is possible to access the computer through internet r Lan without user permission?

Yes as long as the other users computer has been set up to accept remote use.

@guiltyspark.. am MCA degree...Plan to do MCSA 2008..looking for server support job..currently working as Desktop Engineer.

in my office am the only one engineer maintaing everything..so i just learning how to take a machine without their permission

since you are a desktop engineer you can take RDP(mstsc) using the local administrator account of the system which you want to work on, or if its in domain use the domain admin account you use for all sorts of troubleshooting(better to get your account added to the admin group), again you can use system management (server\computer manager) tool to do the administrative tasks. there are many tools available in the market for computer management but i can't think of any think free right now..
